The preneoplastic state in mouse mammary tumorigenesis.

چکیده

It is widely accepted that neoplasms in epithelial organs arise via multiple intermediate stages (1—3). This concept has been extensively developed over the past 30 years and is based upon both clinical and experimental observations in practically all major epithelial organs. The intermediate stages have been labeled with various terms, although the terms preneoplastic or premalignant have been used most frequently. It is the aim of this commentary to discuss the preneoplastic state in the mouse mammary gland. It is hoped that the review will invite discussion and comment, stimulate new ideas and experimental approaches, and attract new participants into this research area. The time is appropriate for such a review in light of the new methodologies which have been developed in the area of mammary gland research over the past 10 years and the opportunity to apply newer molecular biology approaches to examine basic questions in mammary carcinogenesis.

Stroma is not a major target in DMBA-mediated tumorigenesis of mouse mammary preneoplasia.

Carcinogenesis in the mammary gland is thought to involve carcinogen-induced initiation in mammary epithelial cells. Recent experiments have demonstrated that the mammary stroma can be a target of carcinogenic agents, which results in the stroma positively affecting carcinogenesis. To determine whether the stroma or epithelium is the primary target in chemically induced mouse mammary tumorigene...

Radiation-induced tumorigenesis in preneoplastic mouse mammary glands in vivo: significance of p53 status and apoptosis.

In mouse mammary tumorigenesis, p53 mutations facilitate tumorigenesis in concert with other oncogenic alterations. Ionizing radiation enhances tumorigenesis in preneoplastic mammary outgrowth lines and induces p53-dependent apoptosis. We asked if normal p53 function modulates radiation-induced tumorigenesis in preneoplastic mammary lesions by affecting the apoptotic pathway of cell deletion. T...

Immortalized mouse mammary cells in vivo do not exhibit increased telomerase activity.

The acquisition of immortalization is an early and carefully documented event in mouse mammary tumorigenesis. Activation of telomerase activity is one hypothesis to explain the acquisition of immortalization. We examined the activity of telomerase in well-defined immortalized, non-tumor cell populations of mouse mammary tissue in vivo. The results indicated that normal virgin and mid-pregnant m...

Correlation of natural killer activity with tumorigenesis of a preneoplastic mouse mammary lesion.

Tissue infiltrating lymphocytes isolated from the preneoplastic mouse mammary hyperneoplastic alveolar nodule (HAN) tissue line C4 express high levels of natural killer (NK) activity, which gradually wanes as spontaneous tumors develop (W. Z. Wei and G. Heppner. Br. J. Cancer, 55: 589-594, 1987). Experiments were performed to test whether modulation of NK cell activity would be associated with ...

Role of MMTV integration locus cellular genes in breast cancer.

Mouse mammary tumorigenesis as a result of mouse mammary tumor virus (MMTV) integrations has helped to identify a wide variety of interesting genes that play a role in mammary development and tumorigenesis. Several such genes int1/wnt1, wnt3, wnt 10B, int2/fgf3, fgf4, int3/notch and int6 have been shown to be genetically altered in naturally formed mammary tumors as a consequence of MMTV integr...
